The active ingredient present in Avagra tablets is Avanafil. Each Avagra tablet contains 100 mg of Avanafil.
Mechanism of Action
The physiologic mechanism of erection of the penis involves the release of nitric oxide (NO) in the corpus cavernosum during sexual stimulation. NO then activates the enzyme guanylate cyclase, which causes increased levels of cGMP, producing smooth muscle relaxation in the corpus cavernosum and allowing inflow of blood.
Avanafil has no direct relaxant effect on isolated human corpus cavernosum, but enhances the effect of NO by inhibiting PDE5, which is responsible for degradation of cGMP in the corpus cavernosum. Because sexual stimulation is necessary to start the local release of nitric oxide, the inhibition of PDE5 has no effect in the absence of sexual stimulation.

Avagra Tablets – Dosage
The recommended starting dose of Avagra tablets is 100 mg. Avagra should be administered orally as needed as early as approximately 15 minutes before sexual activity. This medication may be taken with or without food.
Based on individual efficacy and tolerability, the dose can be increased to 200 mg taken as early as approximately 15 minutes before sexual activity, or decreased to 50 mg taken approximately 30 minutes before sexual activity.
The lowest dose that provides the expected medical benefit must be used. The maximum recommended dosing frequency of this medicine is once per day. i.e. 24 hours should elapse before you take the next dose. The most common Avagra side effects with an incidence rate greater than or equal to 2% include headache, flushing, nasal congestion, nasopharyngitis, and back pain.

Avagra Tablets – Contraindications
Avagra tablets are contraindicated in patients with a hypersensitivity to Avanafil or any inactive ingredient present in tablets.

Warnings and Precautions to be taken when using Avagra (Generic Avanafil tablets)
- Before using Avagra tablets please inform your doctor about all the medicines that you take including no prescription medications, over the counter medicines and herbal remedies.
- Patients should not use Avagra if sexual activity is inadvisable due to cardiovascular status or any other reason.
- Use of Avagra with alpha-blockers, other antihypertensives, or substantial amounts of alcohol (greater than 3 units) could lead to hypotension.
- Patients should seek emergency treatment if an erection lasts greater than 4 hours. In case this condition is not treated immediately, penile tissue damage and permanent loss of potency could result.
- Patients should stop Avagra and seek medical assistance in case a sudden loss of vision occurs in one or both eyes, which could be a sign of Non Arteritic Ischemic Optic Neuropathy (NAION). Discuss with patients the increased risk of NAION in patients with a history of NAION.
- Patients should stop taking Avagra and seek prompt medical attention in the event of sudden decrease or loss of hearing.
- Patients taking concomitant strong CYP3A4 inhibitors (including Ketoconazole, Ritonavir, Atazanavir, Clarithromycin, Indinavir, Itraconazole, Nefazodone, Nelfinavir, Saquinavir and Telithromycin), must not use Avagra.
- Avagra can potentiate the hypotensive effect of nitrates, alpha-blockers, antihypertensives, and alcohol.
- Doctors should discuss with patients the potential for Avagra to augment the blood pressure-lowering effect of alpha -blockers and other antihypertensive medications.
- Patients should be made aware that both alcohol and PDE5 inhibitors including Avagra act as vasodilators. When vasodilators are taken in combination, blood-pressure-lowering effects of each individual compound may be increased. Therefore, doctors must inform patients that substantial consumption of alcohol (e.g., greater than 3 units) in combination with Avagra tablets could increase the potential for orthostatic signs and symptoms, including increase in heart rate, decrease in standing blood pressure, dizziness, and headache.
- Drinking alcohol can increase certain side effects of Avanafil. Avoid drinking more than 3 alcoholic beverages while you are taking Avanafil.
- Grapefruit and grapefruit juice may interact with Avanafil and lead to potentially dangerous effects. Discuss the use of grapefruit products with your doctor.
- Avoid using other medicines to treat erectile dysfunction, such as Sildenafil, Tadalafil, or Vardenafil while you are taking Avanafil tablets.
- This medicine will not protect you from sexually transmitted diseases–including HIV and AIDS. Using a condom is the only way to protect yourself from these diseases.
Avagra (Generic Avanafil Tablets) – Drug Interactions

Nitrates: Administration of Avagra (Generic Avanafil tablets) to patients who are using any form of organic nitrate, is contraindicated. In a clinical pharmacology trial, Avanafil was shown to potentiate the hypotensive effect of nitrates.
In a patient who has taken Avanafil, where nitrate administration is deemed medically necessary in a life-threatening situation, at least 12 hours should elapse after the last dose of Avanafil before nitrate administration is considered. In such cases, nitrates should only be administered under close medical supervision with appropriate hemodynamic monitoring.
Alpha-Blockers: Caution is advised when PDE5 inhibitors are co-administered with alpha-blockers. PDE5 inhibitors, including Avanafil, and alpha-adrenergic blocking agents are both vasodilators with blood pressure-lowering effects.
Antihypertensives: PDE5 inhibitors, including Avanafil, are mild systemic vasodilators.During a clinical pharmacology trial it was observed that additional reductions in blood pressure of 3 to 5 mmHg occurred following co-administration of a single 200 mg dose of Avanafil with these agents compared with placebo.
Alcohol: Both alcohol and PDE5 inhibitors, including Avanafil, act as vasodilators. When vasodilators are taken in combination, blood pressure-lowering effects of each individual compound may be increased.

Single doses up to 800 mg of Avanafil tablets have been given to healthy subjects, and multiple doses up to 300 mg have been given to patients. In cases of overdose, standard supportive measures should be adopted as required.
