Uses of Tadalafil Tablets
Tadalafil is used to treat male sexual function problems (impotence or erectile dysfunction-ED). In combination with sexual stimulation, tadalafil works by increasing blood flow to the penis to help a man get and keep an erection. Drug Class and Mechanism
Tadalafil (Tadali Pocket Tablets) is a phosphodiesterase inhibitor type 5. It works by helping the blood flow into a penis to achieve and maintain an erection. Penile erection during sexual stimulation is caused by increased penile blood flow resulting from the relaxation of penile arteries and corpus cavernosal smooth muscle. This response is mediated by the release of nitric oxide (NO) from nerve terminals and endothelial cells, which stimulates the synthesis of cGMP in smooth muscle cells.
Cyclic GMP causes smooth muscle relaxation and increased blood flow into the corpus cavernosum. The inhibition of phosphodiesterase type 5 (PDE5) enhances erectile function by increasing the amount of cGMP. Tadalafil inhibits PDE5. Tadali Pocket Contraindications
Tadali Pocket tablets are contraindicated in patients with a hypersensitivity to Tadalafil or any inactive ingredient present in tablets. Tadali Pocket tablets are also contraindicated in the following cases:
Administration of Tadali Pocket (Generic Tadalafil Tablets) to patients using any form of organic nitrate is contraindicated. Tadalafil was shown to potentiate the hypotensive effect of nitrates. During clinical studies, Tadalafil was shown to potentiate the hypotensive effects of both acute and chronic nitrate administration.
Administration of Tadalafil along with guanylate cyclase (GC) stimulators, such as Riociguat is also contraindicated.

Tadalafil is contraindicated in patients who have loss of vision in one eye because of non-arteritic anterior ischaemic optic neuropathy (NAION), regardless of whether this episode was in connection or not with previous PDE5 inhibitor exposure.
Warnings and Precautions when using Tadalafil Tablets
Do not take Tadalafil, if you are also using a nitrate drug for chest pain or heart problems, including
- nitroglycerin (Nitrostat, Nitrolingual, Nitro-Dur, Nitro-Bid, Minitran, Deponit, Transderm-Nitro),
- isosorbide dinitrate (Dilatrate-SR, Isordil, Sorbitrate),
- isosorbide mononitrate (Imdur, ISMO, Monoket), or recreational drugs such as amyl nitrate or nitrite ("poppers").
Patients should not use Tadali Pocket if sex is inadvisable due to cardiovascular status.
Use of Tadali Pocket with alpha blockers, antihypertensives or substantial amounts of alcohol (5 units or more) may lead to hypotension.
If taking potent inhibitors of CYP3A4, dose should be adjusted: Tadalafil for use as needed: Maximum dose of 10 mg every 72 hours. For once daily use: dose not to exceed 2.5 mg.
Patients should seek emergency treatment if an erection lasts for more than 4 hours Use Tadalafil with caution in patients predisposed to priapism.
Patients should stop Tadalafil (Tadali Pocket Tablets) and seek medical care if a sudden loss of vision occurs in one or both eyes, which could be a sign of Non Arteritic Ischemic Optic Neuropathy (NAION). Discuss increased risk of NAION in patients with history of NAION.
Patients should stop Tadali Pocket tablets and seek prompt medical attention in the event of sudden decrease or loss of hearing.
Taking Tadali Pocket (Tadalafil Tablets) with a nitrate medicine can cause a serious decrease in blood pressure, leading to fainting, stroke, or heart attack. Before taking Tadalafil, tell your doctor, if you have:
- heart disease or heart rhythm problems;
- a recent history a heart attack (within the past 90 days);
- a recent history of stroke or congestive heart failure (within the past 6 months);
- angina (chest pain);
- high or low blood pressure;
- liver disease;
- kidney disease (or if you are on dialysis);
- a blood cell disorder such as sickle cell anemia, multiple myeloma, or leukemia;
- a bleeding disorder such as hemophilia;
- a stomach ulcer;
