The active pharmaceutical ingredients contained in Nizonide tablets is Nitazoxanide. Each Nizonide tablet contains 500 mg of Nitazoxanide.
Mechanism of Action
The antiprotozoal activity of Nitazoxanide is believed to be due to interference with the pyruvate:Ferredoxin oxidoreductase (PFOR) enzyme-dependent electron transfer reaction which is essential to anaerobic energy metabolism.
Studies have shown that the PFOR enzyme from Giardia lamblia directly reduces nitazoxanide by transfer of electrons in the absence of ferredoxin. The DNA-derived PFOR protein sequence of Cryptosporidium parvum appears to be similar to that of Giardia lamblia.
Interference with the PFOR enzyme-dependent electron transfer reaction may not be the only pathway by which Nitazoxanide exhibits antiprotozoal activity.
Uses of Nizonide Tablets (Nitazoxanide Generic)
Nitazoxanide Tablets (Nizonide Tablets) treats diarrhea caused by parasite infections. It works by stopping the growth of the parasite. It will not treat colds, the flu, or infections caused by bacteria or viruses.
Nitazoxanide Tablets have not been shown to be effective for the treatment of diarrhea caused by Cryptosporidium parvum in HIV-infected or immunodeficient patients.
Nitazoxanide Tablets (Nizonide Tablets) – Dosage
The recommended dosage of Nitazoxanide Tablets in patients 12 years and older is 500 mg orally every 12 hours with food for 3 days.
Nitazoxanide Tablets should not be administered to pediatric patients 11 years of age or younger because a single tablet contains a greater amount of Nitazoxanide than the recommended dosing in this pediatric age group.
Nitazoxanide Tablets are not interchangeable with Nitazoxanide for Oral Suspension.
Missed Dosage Instructions for Nizonide Tablets
Take the missed dose of Nizonide (Nitazoxanide 500 mg tablets) as soon as you remember.
However, if it is almost time for your next scheduled dose, skip the missed one and continue with your regular dosing schedule. Do not take a double dose to make up for the forgotten one.Taking two doses too close together increases the risk of side effects like nausea, stomach pain, or headaches.
Contraindications
Nizonide tablets are contraindicated in persons with a hypersensitivity (allergy) to Nitazoxanide or any of the other ingredients of this medicine.

The pharmacokinetics of Nitazoxanide in patients with compromised renal or hepatic function have not been studied. Therefore, nitazoxanide must be administered with caution to patients with hepatic and biliary disease, to patients with renal disease and to patients with combined renal and hepatic disease.
Tizoxanide is the primary active metabolite of the broad-spectrum anti-parasitic and antiviral drug Nitazoxanide. Tizoxanide is highly bound to plasma protein (greater than 99.9%). Therefore, caution should be used when administering Nitazoxanide concurrently with other highly plasma protein-bound drugs with narrow therapeutic indices, as competition for binding sites may occur (e.g., Warfarin).
Side Effects of Nitazoxanide Tablets (Nizonide Tablets)
The most common side effects in 2% or more of patients using this medication were abdominal pain, headache, chromaturia, and nausea. The following is a list of side effects spontaneously reported with Nitzoxanide Tablets which were not included in clinical trial listings:
- Gastrointestinal disorders: diarrhea, gastroesophageal reflux disease
- Nervous System disorders: dizziness
- Respiratory, thoracic and mediastinal disorders: dyspnea
- Skin and subcutaneous tissue disorders: rash, urticaria
Overdosage
Information on Nitazoxanide overdosage is not available.
Single oral doses of up to 4000 mg Nitazoxanide have been administered to healthy adult volunteers without significant adverse effects.
In the event of overdose, gastric lavage may be appropriate soon after oral administration. Patients should be carefully observed
and given symptomatic and supportive treatment.
Nitazoxanide Tablets (Nizonide Tablets) During Pregnancy
Nitazoxanide tablets are classified by the US FDA as Pregnancy Category B.
It is not known whether is excreted in human milk.
Reproduction studies have been performed at doses up to 3200 mg/kg/day in rats (approximately 26 times the clinical adult dose adjusted for body surface area) and 100 mg/kg/day in rabbits (approximately 2 times the clinical adult dose adjusted for surface area) and have revealed no evidence of impaired fertility or harm to the fetus due to Nitazoxanide. There are, however, no adequate and well-controlled studies in pregnant women.
It is not known whether Nitazoxanide is excreted in human milk. Because many drugs are excreted in human milk, caution should be exercised when Nitazoxanide is administered to a nursing woman.
